She came to prominence after appearing on the series Mean Girls (2004), and Cloverfield (2008). Afterwards, Caplan went on to star in Related (2005–2006), The Class (2006–2007), Party Down (2009–2010; 2023), and many others TV shows.

In 2013, Caplan began to play the role of Virginia E. Johnson on the Showtime series Masters of Sex,.

The Primetime Emmy Award nominee also portrayed Annie Wilkes in the Hulu anthology series Castle Rock.
