She started her acting career in 1999 when she played the role of a girl named Sara in the tv series Freaks and Geeks.

In 2000, she also appeared in her first film titled From Where I Sit. She played Tina Greer in an episode of Smallville in 2001, as well as two episodes of Once And Again that same year.
Caplan starred in Cloverfield in 2008, Save the Date and Bachelorette and New Girl in 2012, and The Interview in 2014.
In 2013, she starred in the Showtime series, Masters of Sex. She starred in the series until 2016, The Disaster Artist (2017), Extinction (2018), Truth to be Told (2019-2020), Inside Job (2021-2022), and Fatal Attraction (2023).
