Born in Buenos Aires, Cambiasso began his professional career with Argentinos Juniors in 1995, before moving to Real Madrid in 1996 along with his brother Nicolás Cambiasso.
After failing to impress, Esteban moved back to Argentina in 1998, where he played for three years with Independiente and one with River Plate.
After gathering useful experience, he returned to Real Madrid in 2002 and helped Los Blancos win the UEFA Super Cup and the Intercontinental Cup in 2002, the Liga and the Supercopa de España in 2003, and also reached the semi-finals of the UEFA Champions League that season.
Cambiasso also reached the Copa del Rey final the following season.

In July 2004, Cambiasso signed with Serie A club Inter Milan after his contract with Real Madrid expired the previous month.
He stayed at Inter for ten seasons, winning several trophies, including five Scudetti and the 2009–10 UEFA Champions League.
He left Inter Milan at the end of the 2013–14 season, after not being offered a contract extension on its expiration.
After departing Inter Milan, he played for Leicester City and Olympiacos.
He announced his retirement from professional football on 8 September 2017, two months after his contract with Olympiacos expired.
Cambiasso retired winning a total of 21 official trophies.
