Collin Gillespie was born on June 25, 1999, in Huntingdon Valley, Pennsylvania, United States of America. As a senior at Archbishop Wood Catholic High School, Collin Gillespie led the team to a PIAA Class 5A state title in 2017.

Collin Gillespie enrolled at Villanova University in the fall of 2017 and made his first start on February 10, in an 86–75 win over Butler.

As a sophomore at Villanova, Collin Gillespie averaged 10.9 points per game. Gillespie had 27 points and six assists in an 87–78 loss to Baylor in the championship game of the Myrtle Beach Classic.
After going undrafted in the 2022 NBA draft, Collin Gillespie signed a two-way contract with the Denver Nuggets. In the summer of 2019, Collin Gillespie was a part of the United States National team who competed at the Pan American Games in Peru.
