Mitsopoulos began her media career in 1991 with Albany, Western Australia-based Golden West Network. She joined Network Ten in 1993 and went to STW in Perth in 1998 following a period with Associated Press Television News in London.

Mitsopolous spent 13 years with STW, mostly as the station’s state political correspondent.
She began working for the Australian Broadcasting Corporation in 2011.
Mitsopoulos began co-hosting ABC Radio Perth’s weekday Breakfast show with Russell Woolf in January 2019.
In January 2021, she switched to the Mornings program.
Mitsopoulos is a member of Lifeline’s board of directors.
