Amell is the son of Josephine Burden and Robert Patrick Amell III who work in the custom jewelry business. According to sources, he started his career in the entertainment industry as a model when he was just 6 years old.

He made his acting debut at age 16 when he landed several roles in high school in plays such as Louis and Dace and Fiona, Picasso at the Lapin Agile, The Importance of Being Earnest, and more.

After high school, he developed so much for acting and he was influenced to attend the Canadian Studios Acting Academy. He also attended Lawrence Park Collegiate Institute in Toronto and graduated in 2006.

Amell pursued acting as a full-time career after graduating from college in 2006. Some of his earliest movies include American Pie Presents: Beta House, Left for Dead, Picture This, The Alyson Stoner Project, Stuck by Lightning, and more.
