On September 11, 2015, Murphy was promoted to Major League Baseball and made his debut the next day against the Seattle Mariners at Safeco Field. He hit his first MLB home run on September 19 at Coors Field off San Diego pitcher Marcos Mateo.
He was claimed off waivers by the San Francisco Giants on March 25, 2019. He was designated for assignment on March 28, after failing to make the Opening Day roster. The next day, he was traded to the Seattle Mariners for Jesus Ozoria.
In 2020, Murphy missed the entire season staying on the injured list with a fractured metatarsal in his left foot. On January 12, 2023, he agreed to a one-year, $1.625 million contract with the Mariners avoiding salary arbitration.
