In 2021, Brito split the year between the High-A Hudson Valley Renegades and Double-A Somerset Patriots, amassing an impressive record with 22 starts, a 7–7 win-loss record, and a 3.55 ERA. Notably, he recorded 118 strikeouts in 116+2⁄3 innings pitched during this season.

The 2022 season saw Brito showcasing his skills with Somerset and the Triple-A Scranton/Wilkes-Barre RailRiders. He compiled a cumulative record of 11–4 with a 2.96 ERA, registering 91 strikeouts in 112+2⁄3 innings across 26 games, 23 of which were starts.

Recognizing his potential, the New York Yankees selected Brito to the 40-man roster on November 10, 2022, protecting him from the Rule 5 draft. He made his highly anticipated major league debut on April 2, representing a significant milestone in his career. However, on May 20, Brito was optioned to Triple-A Scranton to make room on the roster for Severino’s return.

The latest development in Jhony Brito’s career occurred on December 6, 2023, when the Yankees engaged in a trade with the San Diego Padres. Brito, along with Michael King, Drew Thorpe, Randy Vásquez, and Kyle Higashioka, was part of the package traded to the Padres in exchange for Juan Soto and Trent Grisham.
