Brito’s journey in professional baseball began when he signed with the New York Yankees as an international free agent on November 9, 2015. Making his professional debut in 2016 with the Dominican Summer League Yankees, Brito showcased his talent and dedication to the sport.

The 2022 season saw Brito further solidify his skills, spending time with Somerset and the Triple-A Scranton/Wilkes-Barre RailRiders. He compiled an 11–4 record and a 2.96 ERA with 91 strikeouts in 112+2⁄3 innings across 26 games, including 23 starts. These achievements caught the attention of baseball enthusiasts and experts alike.

On November 10, 2022, recognizing his talent, the Yankees selected Jhony Brito to the 40-man roster to protect him from the Rule 5 draft. His major league debut with the Yankees took place on April 2, marking a significant milestone in his career.

In a notable development on December 6, 2023, the Yankees traded Jhony Brito, along with several other players, to the San Diego Padres in exchange for Juan Soto and Trent Grisham.
